It may be applied when there is a personal or family history of venous thromboembolism, unusual clotting episodes, pregnancy complications, or prior clots associated with hormone therapy, surgery, or immobilization.

The profile commonly includes genetic testing for inherited variants linked to thrombophilia, such as Factor V Leiden (F5 R506Q) and the Prothrombin G20210A mutation, as well as laboratory assessments for deficiencies of natural anticoagulants (protein C, protein S, and antithrombin). Additional tests may cover antiphospholipid antibodies, lupus anticoagulant, anticardiolipin antibodies, and markers of elevated homocysteine. Tests vary by laboratory and clinical indication.
