Instrumentsthermocyclers, commonly referred to as thermocyclers or PCR machines, are laboratory instruments that automate the cycling of temperatures required for polymerase chain reaction (PCR) to amplify DNA. They provide precise temperature control, uniform heat transfer, and programmable cycling protocols, enabling reproducible DNA amplification across many samples.

Most thermocyclers include a temperature-controlled block, Peltier elements for heating and cooling, a heated lid to prevent condensation, temperature sensors for feedback, and software for program design and run control. They may feature gradient capability to test multiple annealing temperatures in a single run and support different formats such as 96-well or 384-well plates and various tube configurations.

Operation and output vary by model. In standard PCR, programs cycle through denaturation, annealing, and extension steps with specific temperatures and times. Typical temperatures are around 95 C for denaturation, 50–65 C for annealing, and 68–72 C for extension, though exact values depend on primers and polymerase. Conventional thermocyclers perform endpoint amplification, while real-time or quantitative PCR (qPCR) thermocyclers monitor fluorescence during amplification to estimate product quantity after each cycle.

Types and configurations include conventional thermocyclers for standard PCR, real-time qPCR systems with integrated optics for fluorescence detection, gradient thermocyclers for temperature optimization, and various throughput formats (plates, tubes) and automation compatibility.

Applications span molecular biology, clinical diagnostics, genetics research, microbial detection, forensics, and food safety testing. Thermocyclers enable cloning, genotyping, mutation screening, expression analysis, and pathogen detection, making them a foundational tool in many laboratories.

Maintenance and quality control involve regular calibration of temperature accuracy and uniformity, verification of lid pressure, cleaning, and software updates to ensure reproducibility and regulatory compliance in clinical settings.
