A prodrugista designs promoieties to improve pharmacokinetic properties, such as oral bioavailability, solubility, tissue targeting, and reduction of toxicity. They work across medicinal chemistry, pharmacology, and drug development teams, performing literature reviews, selecting activation pathways, planning synthetic routes, and assessing stability and activation kinetics both in vitro and in vivo. They use in silico tools and structure-activity data to predict activation and PK properties.

Design strategies and examples

Common strategies include ester, carbonate, phosphate, and amide linkers that mask polar groups; carbohydrate or amino acid promoieties for targeted release; enzyme-triggered activation by esterases, phosphatases, or other enzymes. Notable examples of prodrugs developed with such strategies include capecitabine (prodrug of 5-fluorouracil), valganciclovir (prodrug of ganciclovir), oseltamivir (prodrug of oseltamivir carboxylate), and enalapril (prodrug of enalaprilat).
