Hypertrophy refers to the growth of an organ or tissue due to an increase in the size of its cells, rather than an increase in their number. This process is distinct from hyperplasia, which involves an increase in cell number. Hypertrophy is commonly observed in muscle tissue, where it results in increased muscle mass and strength, a phenomenon often pursued in strength training and bodybuilding.

In skeletal muscles, hypertrophy occurs in response to mechanical stress, such as resistance exercise. When muscles are subjected to progressive overload—gradually increasing the demands placed on them—the body adapts by increasing muscle fiber size. This adaptation is mediated by various biological mechanisms, including the activation of satellite cells, which contribute to muscle repair and growth, and the upregulation of protein synthesis pathways, particularly those involving insulin-like growth factor 1 (IGF-1) and mechanistic target of rapamycin (mTOR).

Hypertrophy can also occur in non-muscle tissues, such as the heart, where it may develop as a compensatory response to increased workload, such as chronic hypertension or valvular heart disease. In these cases, hypertrophy can initially improve cardiac function but may eventually lead to pathological conditions, including heart failure, if the stress persists.

The regulation of hypertrophy involves complex signaling pathways that integrate mechanical, hormonal, and metabolic cues. Growth factors, cytokines, and mechanical stimuli all play roles in modulating the hypertrophic response. While beneficial in controlled contexts like muscle training, excessive or pathological hypertrophy can contribute to disease progression, underscoring the importance of balanced physiological regulation.

Understanding hypertrophy is crucial in fields such as sports science, medicine, and rehabilitation, where optimizing muscle growth or mitigating adverse hypertrophic effects is a key objective.
