Bioprintimine

Bioprintimine is a term used in speculative discussions and early-stage research to describe a class of crosslinking systems for 3D bioprinting. In these approaches, imine chemistry is employed to form covalent bonds between reactive groups on polymers and biomolecules, enabling rapid gelation under aqueous, physiological conditions. Bioprintimine formulations are typically integrated into bioinks to increase print fidelity, mechanical stability, and, potentially, biocompatibility with living cells.

There is no single, standardized compound known as bioprintimine. Rather, the term describes a family of imine-based crosslinkers and dynamic covalent links used in diverse bioink designs. As a concept, it has appeared mainly in exploratory literature and industry discussions rather than as a widely adopted commercial product.

The core mechanism involves formation of imine bonds, or Schiff bases, between aldehyde- or ketone-functional groups and primary amines on polymers such as gelatin, alginate, or amino-modified biopolymers. These bonds can be designed to be reversible or dynamic, allowing remodeling of the hydrogel network and tunable stiffness. Reaction conditions are typically mild, compatible with cells, and adjustable through ligand structure and pH.

Applications include tissue-like hydrogels for tissue engineering, organ-on-a-chip models, and cell-laden constructs where rapid gelation, good print resolution, and minimal cytotoxicity are valued. Bioprintimine-based bioinks aim to support high cell viability during printing and allow post-print maturation.

Challenges include potential cytotoxicity from aldehyde-containing components, control of degradation and swelling, batch-to-batch variability, and regulatory uncertainties for new chemical crosslinkers.
