Prolinerich is an adjective used in molecular biology to describe proteins, regions, or sequences with a high proportion of the amino acid proline. The term emphasizes the abundance of proline residues, which can influence structure, dynamics, and interactions. Prolinerich regions are often found in intrinsically disordered regions and low-complexity domains and may be defined quantitatively by a proline content threshold (for example, exceeding 20–30% proline) or by prevalence of proline-rich motifs such as PxxP.

Structural properties: Proline imposes conformational constraints that affect folding. Proline-rich segments frequently adopt extended polyproline II helices and can disrupt alpha helices and beta sheets. The presence of PxxP motifs provides docking sites for Src homology 3 (SH3) domains and other modules, enabling transient protein-protein interactions.
