Conformational refers to the arrangement of atoms in a molecule that can be interconverted into other arrangements by rotations about single bonds, without breaking any bonds. In chemistry, this term distinguishes conformational isomerism from configurational isomerism; conformers (or rotamers) interconvert rapidly at room temperature in many molecules, while configurational isomers require bond breaking to interconvert. The study of past and present conformations is known as conformational analysis.

Classic examples include ethane, which has staggered and eclipsed conformations around the C–C bond; staggered is lower in energy due to reduced torsional strain. Butane exhibits anti and gauche conformations; the gauche interaction raises energy relative to anti. Rings behave differently: cyclohexane adopts a chair conformation that minimizes steric strain and rapidly undergoes ring flips to a boat or twist-boat form under certain conditions; in many cases the chair is most stable. Some molecules display restricted rotation due to steric hindrance or partial double-bond character, limiting the accessible conformations.

Conformational analysis uses experimental data and computational methods to map a molecule’s conformational landscape, estimate populations via Boltzmann distribution, and predict properties such as reactivity and binding affinity. Techniques include NMR spectroscopy, X-ray crystallography, and computational chemistry (molecular mechanics and quantum calculations). Conformational considerations are crucial in biochemistry, where proteins and nucleic acids undergo conformational changes that underpin function, and in materials science, where polymer conformation affects mechanical and optical properties.
