Odealkylation, also referred to as O-dealkylation, is a chemical process in which an alkyl group bound to an oxygen atom in an ether is removed. The reaction typically yields an alcohol or phenol and a carbonyl-containing fragment (an aldehyde or ketone) corresponding to the alkyl group that was cleaved. In biochemistry, odealkylation most often describes oxidative metabolism of aryl-alkyl ethers, particularly O-demethylation when the departing group is a methyl, and more generally O-dealkylation for longer alkyl substituents.

In biological systems, odealkylation is commonly catalyzed by cytochrome P450 enzymes. The mechanism involves activation of the C-O bond, often by hydrogen abstraction and oxygen insertion, followed by cleavage of the bond to release the alkyl fragment as an aldehyde or related oxidized product, while the remaining oxygen-containing fragment becomes a phenol or alcohol. For example, O-demethylation of anisole yields phenol and formaldehyde, whereas O-deethylation can yield phenol and acetaldehyde.

In synthetic chemistry, O-dealkylation refers to the cleavage of ethers to remove an alkyl group, frequently under acidic or oxidative conditions. Typical methods include treatment with strong acids (which can yield alkyl halides alongside phenols) and reagents such as boron tribromide for aryl-alkyl ethers. The choice of method depends on substrate structure and the desired products.

Applications and significance include drug metabolism, where O-dealkylation can deactivate or activate compounds and influence pharmacokinetics, and protective-group strategies in organic synthesis, where methoxy or alkoxy groups are removed to reveal phenols. Factors such as electronic effects and sterics govern rate and selectivity.
