Craniofaciallimb is a descriptive term used in medical genetics and dysmorphology to denote congenital conditions in which craniofacial structures and limb development are affected simultaneously. It is not a single disease but an umbrella classification applied to multiple syndromes and malformation patterns that show concurrent head/face anomalies and limb abnormalities.

Clinical features are variable and depend on the underlying cause, but typically involve a combination of craniofacial signs (such as facial asymmetry, midface hypoplasia, ear or eye anomalies, or clefting in some cases) together with limb findings (including limb reduction or hypoplasia, digit abnormalities, or limb length discrepancies). The spectrum ranges from mild to complex malformations, and associated conditions can include sensory, neurologic, or cardiovascular findings.

Etiology is diverse and often genetic, involving disruptions in early embryonic development that influence neural crest cell migration, facial patterning, or limb bud formation. Mutations in various developmental genes and signaling pathways are implicated, and environmental factors may contribute in some instances. Many cases are sporadic, while others follow recognizable inheritance patterns.

Diagnosis relies on thorough clinical assessment supported by imaging studies and, when available, genetic testing such as gene panels, microarray, or exome sequencing. A multidisciplinary approach is typically required, involving plastic surgery or maxillofacial surgery, orthopedics, physical and occupational therapy, audiology or ophthalmology as indicated, and genetic counseling.

Prognosis varies with the specific condition and the severity of craniofacial and limb involvement. Management focuses on optimizing function and appearance, enabling development, and addressing associated health issues through coordinated care.
