Melanocyte-related refers to anything pertaining to melanocytes, the pigment-producing cells of the skin, hair, and eyes. Melanocytes originate from neural crest cells and migrate to the epidermis and hair follicles during development. They synthesize melanin in specialized organelles called melanosomes and transfer melanin-containing packages to neighboring keratinocytes, contributing to skin color and photoprotection.

Melanin production is influenced by genetic and environmental factors. The key enzyme tyrosinase catalyzes the first steps of melanin synthesis, with additional enzymes such as TYRP1 and DCT shaping pigment type. The signaling axis involving the melanocortin 1 receptor (MC1R), alpha-melanocyte-stimulating hormone (alpha-MSH), and the transcription factor MITF controls melanocyte activity. Melanin exists mainly as eumelanin (brown-black) and pheomelanin (red-yellow); their relative amounts affect coloration and UV protection.

Development and distribution: melanocytes populate the epidermis and hair follicles in a site-specific manner, with variation among individuals and body sites. In hair follicles, melanocyte stem cells stored in the bulge region contribute to ongoing hair pigmentation.

Clinical relevance: abnormal melanocyte function or number can lead to pigmentary disorders. Vitiligo involves loss of melanocytes; albinism results from tyrosinase or transport defects impairing melanin production. Hyperpigmentation can follow inflammation or sun exposure. Melanocytic nevi (moles) are benign proliferations, while melanoma is a malignant tumor of melanocytes. Diagnosis uses clinical examination, Wood’s lamp examination, and dermoscopy; treatment ranges from sun protection and topical agents for pigmentary conditions to surgical excision, targeted therapy, or immunotherapy for melanoma.

Research and applications include advances in melanocyte biology, pigmentary disorder management, and melanoma therapies.
