Homeobox-related refers to genes and proteins that carry a homeobox or a closely related DNA-binding domain and encode transcription factors involved in development. The term covers both canonical homeobox genes and additional genes with related sequences that regulate when and where other genes are turned on during embryogenesis and tissue formation.

Most homeobox-related proteins contain a conserved 60-amino-acid homeodomain that binds DNA, often accompanied by other motifs that modulate DNA interaction, localization, or protein partnerships. They are grouped into families by sequence similarity in the homeodomain and surrounding regions, such as HOX, SIX, NK-like, and MEIS families, among others.

Evolutionarily, the homeobox domain is ancient and highly conserved across diverse life forms. In animals, clusters of homeobox genes have undergone duplications and rearrangements that contribute to complex regulatory networks governing body plan and organ development.

Functionally, homeobox-related transcription factors control cell fate, limb and neural patterning, axis specification, and organogenesis. Disruptions in their expression can lead to congenital anomalies and have been linked to various cancers and diseases through misregulation of developmental programs.

In research, these genes are studied in a range of model organisms to understand developmental biology and evolution. They are also explored in regenerative medicine and cancer biology as regulators of cell identity and differentiation.
