XLHED

XLHED, or X-linked Hypohidrotic Ectodermal Dysplasia, is a genetic disorder characterized by the abnormal development of ectodermal structures, including sweat glands, hair, teeth, and sweat production. It is typically inherited in an X-linked pattern, meaning the defective gene responsible is located on the X chromosome, primarily affecting males, with females often being carriers who may exhibit mild symptoms.

Individuals with XLHED commonly present with reduced or absent sweating (hypohidrosis or anhidrosis), leading to difficulties in temperature regulation and an increased risk of overheating. Other hallmark features include fine or sparse hair (hypotrichosis), missing or cone-shaped teeth (hypodontia or oligodontia), dry skin, and characteristic facial features such as a prominent forehead, thick or rudimentary eyebrows and eyelashes, and a flat nasal bridge.

The disorder results from mutations in the EDA gene, which encodes a protein essential for the development and maintenance of ectodermal structures. The severity of symptoms varies among individuals, and the condition may be diagnosed based on clinical evaluation, family history, and genetic testing.

Management of XLHED involves symptomatic treatment, including specialized dental care, skin hydration, and environmental modifications to prevent overheating. Recent advances include potential gene therapy approaches aimed at correcting the underlying genetic defect.

Although XLHED is a lifelong condition, early diagnosis and appropriate healthcare interventions can significantly improve quality of life. Research continues to explore targeted therapies that address the molecular basis of the disorder, with the goal of improving outcomes for affected individuals.
