UCP1 protein, also known as thermogenin, is a mitochondrial inner membrane protein that uncouples oxidative phosphorylation from ATP production in brown adipose tissue. It is encoded by the UCP1 gene and belongs to the mitochondrial carrier protein family. In humans, UCP1 is predominantly expressed in brown adipocytes, though trace expression can be found in other tissues and depots.

The primary function of UCP1 is to dissipate the proton gradient across the inner mitochondrial membrane as heat, rather than driving ATP synthesis. Protons re-enter the matrix through UCP1, releasing energy as heat in a process called non-shivering thermogenesis. Fatty acids act as activators of UCP1, while purine nucleotides such as GDP and ATP inhibit its activity. Cold exposure and sympathetic nervous system signaling upregulate UCP1 expression and promote thermogenic activity, contributing to increased energy expenditure.

Regulation of UCP1 involves transcriptional and signaling pathways, including PGC-1α and PRDM16, which drive mitochondrial biogenesis and brown-fat differentiation. UCP1 expression is higher in infancy and tends to decline with age; it is influenced by nutritional status and adiposity, with obesity often associated with reduced BAT activity and UCP1 levels. In adults, functional brown adipose tissue and detectable UCP1 activity are typically localized to regions such as the neck and upper back, though individual variation is common.

Clinical and research relevance centers on UCP1’s role in adaptive thermogenesis and metabolic rate. Enhanced UCP1 activity or expression is a focus of obesity and metabolic syndrome research, with ongoing studies evaluating therapeutic strategies to stimulate brown-fat thermogenesis. Variants in UCP1 have been investigated for associations with body composition and lipid metabolism, though findings are not yet conclusive.
