Platelet-driven is a term used to describe biological processes in which platelets actively determine outcomes, rather than merely participating. In mammals, platelets respond to vascular injury by rapid activation and aggregation, initiating hemostasis. Beyond stopping bleeding, platelets influence wound healing, angiogenesis, and tissue remodeling through cargo released from alpha and dense granules and by shedding extracellular vesicles.

Platelet-driven processes depend on receptors such as GPVI and GPIb-IX-V for sensing damaged endothelium, and on signaling via P2Y12 and thromboxane receptors that amplify activation. Platelets release growth factors including PDGF, VEGF, and TGF-β, and present adhesion molecules like P-selectin that coordinate interactions with neutrophils, monocytes, and endothelial cells.

In disease, platelets can promote thrombosis, cancer metastasis, and inflammatory states by sustaining platelet-leukocyte alliances and remodeling the vascular and tissue microenvironment. Platelet-derived extracellular vesicles convey procoagulant phospholipids and signaling cues that extend platelet influence systemically.

Therapies targeting platelet-driven pathways, notably antiplatelet drugs, are central to cardiovascular risk management. Research into platelet-driven biology seeks to map how platelets orchestrate hemostasis, immunity, repair, and pathology, enabling more precise interventions and biomarkers.
