Nucleocytoskeletal refers to the integrated network that links the nucleus to the cytoskeleton, coordinating nuclear structure, positioning, and signaling. It encompasses the nuclear envelope, the nuclear lamina, and the cytoskeletal filaments that interface with the envelope to transmit mechanical cues and influence genome organization.

Key structural elements include the nuclear lamina, composed of lamins A/C and B, along with lamin-associated proteins, and the LINC complex (linker of nucleoskeleton and cytoskeleton) that spans the inner and outer nuclear membranes. SUN domain proteins reside in the inner nuclear membrane and bind lamins, while KASH domain proteins extend into the cytoplasm and connect to actin filaments, microtubules, or intermediate filaments via adapters such as nesprins.

Functions of the nucleocytoskeletal system include providing mechanical stability to the nucleus, shaping nuclear morphology, and positioning the nucleus within the cell. It transduces mechanical signals from the cytoskeleton to the genome, influencing chromatin organization and gene expression through lamin-associated domains and changes in higher-order chromatin structure. The network also participates in nuclear migration during development, cell polarization, and the orchestration of nuclear envelope breakdown and reassembly during mitosis.

Dynamic regulation involves forces transmitted through the LINC complex, rearrangements of actin and microtubules that modulate connections, and post-translational modifications of lamins and LINC components in response to signaling and extracellular matrix stiffness. Mechanical context and cell state can thus shape nuclear mechanics and transcriptional programs.

Clinical relevance is found in laminopathies and related disorders caused by defects in nucleocytoskeletal components, including Emery-Dreifuss muscular dystrophy, Dunnigan-type familial partial lipodystrophy, and various cardiomyopathies. Mutations in nesprins or SUN proteins can disrupt nuclear positioning and cell migration, contributing to neurological and developmental phenotypes.

Research approaches include advanced imaging, such as super-resolution and electron microscopy, biochemical reconstitution of LINC complexes, genetic models to dissect function, and mechanical assays to probe nuclear resilience.
