Keratinolysis

Keratinolysis is the enzymatic or chemical breakdown of keratin, the fibrous structural protein that forms hair, nails, and the outer skin layers. In biological contexts, keratinolysis is primarily driven by keratinases—proteases produced by certain microorganisms including dermatophytes such as Trichophyton and Microsporum, as well as various bacteria and actinomycetes. These organisms secrete keratinases and supplementary proteases that cleave keratin’s peptide bonds; the high cysteine content and disulfide cross-links of keratin make degradation difficult, so effective keratinolysis often involves reductive steps that disrupt disulfide bonds and enhance proteolysis.

In medical microbiology, keratinolysis is a key step enabling invasion of keratinized tissues, contributing to diseases such as tinea and onychomycosis. Laboratory assessment of keratinolytic activity—using keratin-containing substrates and observing zones of degradation—helps characterize dermatophytes and related microorganisms.

Industrial and environmental applications exploit keratinases to convert keratin-rich waste (feathers, hair) into value-added products, and to assist in processes such as leather dehairing and bioconversion of keratin-containing materials for animal feed or composting.

Keratinolysis is sometimes contrasted with keratolysis, the broader dermatologic term for shedding of the stratum corneum, which may be chemical or mechanical. The two terms overlap in casual usage but denote distinct mechanisms: enzymatic degradation versus exfoliation.
