The signal is produced when a fluorophore transitions from an excited electronic state back to a lower energy state, typically emitting a photon with a longer wavelength than the exciting light. The resulting emission is characterized by a specific spectral profile, known as the emission spectrum, and is measured as intensity.

The strength and reliability of a fluorescence signal depend on several factors. The intrinsic properties of the fluorophore, such as its quantum yield and extinction coefficient, determine how efficiently it converts absorbed light into emitted light. The concentration of fluorophore, local chemical environment (pH, polarity), and potential quenching species influence signal intensity. Photobleaching, the gradual loss of fluorescence upon prolonged illumination, and autofluorescence from the sample or substrate can affect signal-to-noise ratio. Instrumental factors, including illumination power, detector sensitivity, and the use of appropriate excitation and emission filters or monochromators, also play critical roles.

Fluorescence signals are widely used in biology and chemistry for labeling and detecting molecules. Common applications include fluorescence microscopy, flow cytometry, and quantitative assays, where signals may be displayed as images, spectra, or histograms. Proper controls, calibration, and spectral separation are essential to distinguish specific signals from background and to enable meaningful comparisons across samples.
