Borondipyrromethene

Borondipyrromethene, commonly referred to as the BODIPY core, is the boron-containing dipyrromethene motif that underpins a large family of fluorescent dyes. The core consists of a dipyrromethene framework coordinated to boron with two fluoride ligands, forming a rigid, highly conjugated system. In typical preparations, two pyrrole units condense with an aldehyde to yield the dipyrromethene, which is then complexed with boron difluoride to give the BODIPY structure.

Photophysical properties of BODIPY dyes include strong absorption in the visible region, bright fluorescence with sharp, well-defined emission bands, high molar extinction coefficients, and substantial photostability. The spectra are highly tunable through substituents at the pyrrolic rings or at the meso position, allowing shifts in absorption and emission wavelengths as well as changes in quantum yield and solubility. Substituents can render the dyes more water-soluble or more lipophilic, and can introduce reactive handles for bioconjugation. The core is generally chemically robust and exhibits good pH stability, although specific substitutions may influence sensitivity to environmental factors.

Synthesis and functionalization are straightforward, contributing to the popularity of BODIPY dyes in research and industry. Their versatility, combined with favorable photophysical properties, makes them widely used in fluorescence imaging, biological sensing, photodynamic therapy, and as components in organic electronics and laser dye applications. Ongoing development focuses on extending emission into the red or near-infrared and enhancing water compatibility for in vivo use.
