Receptor-associated describes molecules that interact with cell-surface or intracellular receptors and influence their behavior. These interactions can modulate receptor activation, signal transduction, trafficking to the plasma membrane or endosomes, and receptor turnover.

Categories of receptor-associated proteins include adaptor and scaffold proteins that assemble signaling complexes; co-receptors that enhance or modulate ligand binding; chaperones that assist in receptor folding and maturation; and kinases or phosphatases that chemically modify receptors. Examples include SH2- and PTB-domain proteins that bind phosphorylated receptors; PDZ-domain scaffold proteins that organize receptor complexes; and clathrin adaptor proteins such as AP-2 that help internalization.

A well-studied specific case is the receptor-associated protein (RAP), a chaperone for members of the LDL receptor family. RAP binds to the receptor’s ligand-binding domain in the endoplasmic reticulum, helping proper folding and preventing premature ligand interaction. RAP is typically displaced as receptors mature and traffic through the secretory pathway, allowing normal ligand binding in later compartments.

Functions of receptor-associated factors extend to stabilizing receptors at the membrane, shaping signaling specificity, and directing intracellular routing toward recycling or degradation. In signaling, adaptor and scaffold proteins can determine which pathways are activated by a given receptor.

In research, identifying receptor-associated interactions relies on co-immunoprecipitation, pull-down assays, yeast two-hybrid screens, and proximity ligation. Understanding these interactions contributes to insights into immune signaling, neural development, and metabolic regulation, and may reveal targets for therapeutic intervention when receptor trafficking or signaling is dysregulated.
