Nonintegrating is an adjective used to describe a molecule, construct, or process that does not become permanently integrated into a host genome or larger system. In molecular biology and gene therapy, nonintegrating refers to DNA or vectors that persist as extrachromosomal elements rather than being incorporated into chromosomal DNA.

In gene therapy and virology, several nonintegrating approaches exist. Integrase-deficient lentiviral vectors deliver episomal DNA that can express a transgene without stable integration. Adeno-associated virus vectors often persist as episomes in the nucleus, enabling long-term expression in non-dividing cells while largely avoiding integration. Nonintegrating plasmids and nonintegrating viral vectors may use genetic modifications, absence of integration enzymes, or host factors to prevent chromosomal insertion. Expression from nonintegrating DNA is typically maintained in non-dividing cells and can be diluted in proliferating cells.

Advantages of nonintegrating strategies include a reduced risk of insertional mutagenesis and a more favorable safety profile, making them attractive for vaccines, transient expression studies, and therapeutic settings where temporary expression suffices. They also allow rapid assessment of gene function without permanent genome modification.

Limitations include transient expression in dividing tissues and, for many vectors, limited duration and payload capacity. Expression durability varies by cell type and vector, and re-dosing may be complicated by immune responses. Nonintegrating approaches are often balanced against integrating methods that provide durable expression but pose higher genomic risks, depending on the research or clinical objective.
