Luciferase reporter assays are a class of molecular biology techniques used to study gene expression and regulatory element activity. In a typical assay, a promoter or other regulatory sequence of interest drives the expression of a luciferase reporter gene. The encoded luciferase enzyme catalyzes a light-emitting reaction when its substrate is provided, and the emitted light is quantified with a luminometer. The luminescence intensity reflects the activity of the regulatory element and the transcriptional activity in the tested cells.
