isobologrammer

Isobologrammer is a nonstandard term used to describe a person or software that constructs isobolograms to analyze interactions between two or more agents. The label is informal and is not widely used as a formal profession.

In practice, an isobologrammer may be a pharmacologist, biostatistician, data scientist, or a software tool that specializes in dose–response analysis and combination testing. Such practitioners often work in drug development, toxicology, or pharmacology research and may apply computational methods to generate and interpret isobolograms.

The typical workflow involves designing combination experiments, fitting dose–response curves for each agent, and selecting a fixed effect level (such as the ED50 or EC50). The corresponding combination doses are then plotted on an isobologram, with one axis representing the dose of one agent and the other axis representing the dose of the second agent. The line of additivity (often the Loewe additivity line) denotes combinations expected to be additive; points below this line indicate synergy, while points above indicate antagonism. Proper interpretation requires data quality and attention to model assumptions, as deviations can arise from experimental variability or non-orthogonal dose relationships.

Applications include evaluating potential drug interactions in combination therapies, optimizing therapeutic regimens, and informing toxicology risk assessments. Limitations include dependence on the chosen additivity model, potential misinterpretation when more than two agents are involved, and sensitivity to experimental noise. The concept remains a key analytical approach in pharmacology for assessing whether combining agents yields improved efficacy beyond independent effects.
