Glutationiperoksidaasia, commonly known as glutathione peroxidase (GPx), is a family of selenium-dependent enzymes that play a crucial role in cellular defense mechanisms. These enzymes catalyze the reduction of hydrogen peroxide and organic hydroperoxides into non-toxic compounds, such as water and alcohols, respectively. By neutralizing reactive oxygen species (ROS), glutathione peroxidase helps protect cells from oxidative damage, which can lead to mutations, inflammation, and disease progression.

The primary function of glutathione peroxidase involves the oxidation of glutathione (GSH), a tripeptide composed of glutamate, cysteine, and glycine, which acts as a cofactor. In this reaction, two molecules of GSH are oxidized to form glutathione disulfide (GSH2), while the hydroperoxide substrate is reduced. This process is essential for maintaining redox homeostasis within cells, particularly in the presence of oxidative stress.

Glutathione peroxidase exists in multiple isoforms, each with distinct tissue distributions and substrate specificities. The most well-studied isoform, GPx1, is ubiquitously expressed and primarily targets hydrogen peroxide. Other isoforms, such as GPx2 (intestinal), GPx3 (plasma), GPx4 (phospholipid hydroperoxide glutathione peroxidase), and GPx6 (olfactory), exhibit specialized roles in different tissues and cellular compartments. GPx4, for instance, is particularly important for protecting cellular membranes from lipid peroxidation.

Deficiencies in glutathione peroxidase activity have been linked to various pathological conditions, including neurodegenerative diseases, cardiovascular disorders, and cancer. Conversely, increased expression or activity of these enzymes is often associated with enhanced resistance to oxidative stress. Selenium, an essential trace element, is a critical component of the active site of glutathione peroxidase, and its adequate dietary intake is necessary for optimal enzyme function.
