Glutaredoxiner is not a widely recognized term in standard biochemistry or molecular biology. The canonical name for this class of enzymes is glutaredoxin (plural glutaredoxins). In some texts, “glutaredoxiner” may reflect a typographical error or a non-English variant, and its use should be corroborated with reliable sources.

Glutaredoxins are small redox enzymes that catalyze thiol-disulfide exchange reactions using glutathione (GSH) as a cofactor. They help maintain cellular redox balance by reducing protein disulfides and deglutathionylating mixed disulfides formed between cysteine residues and glutathione. Through these reactions, they participate in redox signaling and protect proteins from oxidative damage. The catalytic cycle typically involves cysteine residues in the active site and a connection to the cellular GSH pool, allowing transfer of reducing equivalents.

There are two main functional classes of glutaredoxins. Dithiol glutaredoxins possess two catalytic cysteines that participate directly in disulfide exchange, while monothiol glutaredoxins rely on a single catalytic cysteine and often function in coordination with glutathione to catalyze reactions. Structural diversity exists across bacteria, archaea, and eukaryotes, with variations in subcellular localization, including cytosol, mitochondria, and chloroplasts, reflecting specialized redox roles.

Distribution and roles extend beyond simple disulfide reduction. Some glutaredoxins contribute to iron-sulfur (Fe-S) cluster assembly and maintenance, notably in mitochondria. In organisms, they are involved in responses to oxidative stress, and their activity can influence processes linked to aging, cancer biology, and neurodegenerative diseases. Because of their central role in redox homeostasis, glutaredoxins are studied as potential targets for therapeutic intervention and as markers of cellular oxidative status.
