Barriers can include epithelial or endothelial linings such as the skin, mucosal surfaces, the intestinal tract, and the blood–brain barrier, among others. The term encompasses elements that contribute to barrier integrity, selective permeability, and defense against environmental challenges.

Barrierassociated components include proteins and structures that compose or regulate barrier junctions, as well as transporters and immune factors operating at barrier sites. Examples include tight junction proteins (claudins, occludin, ZO-1), adherens junction molecules (E-cadherin), and junctional adhesion molecules. Barrierassociated transporters and efflux pumps influence movement of solutes and xenobiotics. Immune mediators such as secretory immunoglobulin A at mucosal surfaces can also be described as barrierassociated because of their role in barrier defense.

In research, barrierassociated status is typically inferred from localization to barrier structures, co-localization with known barrier components, and functional assays that measure barrier integrity, such as transepithelial electrical resistance or permeability to tracers. The concept helps categorize how components contribute to barrier maintenance, permeability regulation, and host defense, as well as how barrier dysfunction may influence disease processes.
