Acetylating is the chemical process of introducing an acetyl group, CH3-CO-, into a molecule. This transfer typically occurs from an acetyl donor to a nucleophilic site such as an amine, alcohol, or thiol, yielding amide, ester, or thioester products. Acetylation is widely used to modify reactivity, properties, or biological recognition and can serve both synthetic and regulatory functions.

In organic synthesis, common acetylating agents include acetic anhydride and acetyl chloride, as well as derivatives like acetyl imidazole. Substrates include amines, which form amides such as acetamides, and alcohols or phenols, which form esters such as acetates. Acetylation can serve as a protecting group strategy to mask reactive functionalities during multi-step sequences; deprotection then regenerates the original functionality under controlled conditions. Reactions are typically carried out under relatively mild conditions with bases or catalysts to facilitate acyl transfer.

In biology, acetylation is often enzymatic, mediated by acetyltransferases that use acetyl coenzyme A (acetyl-CoA) as the acetyl donor. Histone acetylation, a well-studied example, modifies lysine residues and can alter chromatin structure and gene expression, generally promoting transcription. Many other proteins and small molecules undergo acetylation, influencing activity, localization, stability, or interactions. Reversible deacetylation by deacetylases completes the regulatory cycle.

Safety and environmental considerations include the corrosive or irritant nature of many acetylating agents and the generation of acidic byproducts. Reactions should be conducted with appropriate engineering controls, personal protective equipment, and proper waste handling.
