Thicksection

Thicksection, often written as "thick section," is a descriptive term used in microscopy, histology and materials science to refer to a cross-sectional slice that is thicker than standard preparatory sections used for routine analysis. The exact thickness varies by field, but thick sections are generally tens to hundreds of micrometers.

In histology and neuroscience, thick sections preserve more of the tissue's three-dimensional structure and connectivity, enabling better visualization of spatial relationships and for 3D reconstructions. They are commonly produced with vibrating microtomes or similar instruments and may require specialized staining or optical clearing to enhance contrast and penetration.

In materials science and engineering, thick sections allow examination of internal microstructures, grain boundaries, and defects that may be obscured in thinner cuts. Preparation typically involves meticulous grinding and polishing to a relatively thick, flat surface, followed by etching or imaging with light or electron microscopy, or non-destructive methods such as X-ray computed tomography.

Imaging and analysis of thick sections often rely on techniques designed to handle increased light scattering and reduced penetration, including optical clearing, confocal or two-photon microscopy, and reconstructed 3D imaging from serial sections. The term "thick section" thus denotes a size category rather than a single standardized protocol.
