SH2domen

SH2domen is a protein domain class related to the classical SH2 (Src Homology 2) domain, identified as a variant that recognizes phosphotyrosine-containing motifs in signaling proteins. It occurs as a modular component within several multi-domain signaling proteins and in certain adaptor proteins, contributing to the assembly of signaling complexes at sites of phosphorylation.

The structural core of SH2domen is described as similar to other SH2-like modules, typically comprising a central beta-sheet scaffold flanked by alpha-helices. It forms a shallow, positively charged binding pocket that coordinates the phosphate group of phosphotyrosine. In addition to the pY pocket, SH2domen often features loop regions that influence specificity and affinity for particular phosphopeptide contexts, enabling selective interactions with distinct target motifs.

Binding and specificity are determined by residues lining the pY pocket and adjoining determinants, which read the sequence context surrounding the phosphorylated tyrosine.
