In prokaryotes, secretion often relies on specialized pathways to export polypeptides across the cell envelope. The Sec and Tat systems direct proteins to the plasma membrane or periplasm, while other systems (Type I–VI secretion systems) can transport effectors directly outside or into a host cell. Bacterial secreted polypeptides include enzymes, toxins, and siderophores.

Quality control and regulation influence polypeptidesecreting. Cellular signaling controls transcription, translation, folding, and trafficking; misfolded polypeptides trigger unfolded protein response. In biotechnology, secretory expression hosts, such as yeast or mammalian cells, exploit signal peptides to enhance yield of recombinant polypeptides.
