Lipidladdade

Lipidladdade is a Swedish term used to describe molecules, particles, or structures that carry lipid groups or incorporate lipids as part of their composition. The concept is common in biochemistry, molecular biology, and nanomedicine, where lipid association influences localization, interactions, and function.

In biochemistry, lipidladdade proteins or peptides have undergone lipidation, a covalent attachment of lipid moieties to specific amino acid residues. This post-translational modification increases hydrophobicity and promotes membrane association, subcellular targeting, and regulation of signaling pathways. Major forms of lipidation include N-myristoylation, S-palmitoylation, S-prenylation (such as farnesylation and geranylgeranylation), and the addition of glycosylphosphatidylinositol (GPI) anchors. Lipidladdade proteins often show altered localization, stability, and interaction networks, with important roles in processes ranging from signal transduction to trafficking.

In nanomedicine and drug delivery, lipidladdade formulations refer to particles whose surfaces are coated with or composed of lipid components, enabling encapsulation or association of therapeutic cargo. Lipid nanoparticles and liposomes are prominent examples, used to deliver nucleic acids, small molecules, or proteins. Lipid loading affects properties such as stability, cellular uptake, biodistribution, and release kinetics, and has been instrumental in technologies including mRNA vaccines.

Detection and study of lipidation or lipid loading employ methods such as mass spectrometry, metabolic labeling with lipid precursors, acyl-biotin exchange, and click chemistry to track lipid-associated species.
