Hyperdiploidy

Hyperdiploidy is a chromosomal condition in which an organism has more chromosomes than the normal diploid number for that species. In humans, the typical diploid count is 46 chromosomes (23 pairs). A hyperdiploid cell thus contains more than 46 chromosomes, often ranging from 47 to several dozen extra chromosomes. This state can arise through errors in cell division, such as nondisjunction during meiosis or mitosis, leading to the duplication of entire chromosome sets or partial chromosomal segments.

The phenomenon is frequently observed in malignant diseases. Pediatric acute lymphoblastic leukemia (ALL) is one of the most common contexts in which hyperdiploidy is identified; tumors in this disease often contain a chromosome count between 48 and 60, and this karyotypic pattern is associated with a favorable prognosis. Hyperdiploid karyotypes are also seen in various solid tumors, including certain types of colorectal, breast, and ovarian cancers, where they may reflect genomic instability or an adaptive growth advantage.

Outside of cancer, hyperdiploidy can occur as a result of chromosomal mosaicism or in developmental disorders, although these cases are less common and may have varied clinical impacts. Some non-malignant hyperdiploid cell populations are maintained without overt disease, suggesting that the cellular environment and additional genetic alterations influence pathogenicity.

Detection of hyperdiploidy is typically performed through cytogenetic techniques such as karyotyping, fluorescent in situ hybridization (FISH), or high-resolution array comparative genomic hybridization. Advances in next-generation sequencing now allow rapid determination of chromosome numbers and the specific loci involved in copy number changes. Understanding the presence and pattern of hyperdiploidy contributes to risk stratification, treatment selection, and prognostication in oncology, while also informing research into genomic instability mechanisms.
