GFP-expressing refers to cells, organisms, or systems engineered to produce green fluorescent protein (GFP), typically used as a reporter or fusion tag to visualize gene expression, protein localization, or cellular processes. GFP is a fluorescent protein originally isolated from the jellyfish Aequorea victoria. When properly folded, it emits green light upon blue excitation, enabling live imaging of biological events without the need for external substrates.

Expression is achieved by introducing a DNA construct that places the GFP gene under a promoter of choice. In prokaryotes and eukaryotes, GFP can be expressed transiently or stably, and it can be used alone or fused to another protein to monitor localization, trafficking, or interactions. Promoters determine tissue specificity and inducibility, allowing researchers to study dynamic changes in expression.

Detection and measurement rely on fluorescence techniques such as fluorescence microscopy, flow cytometry, or plate readers. GFP is excited near 488 nm and emits around 509 nm, with signal strength reflecting expression levels. Considerations include maturation time, photobleaching, pH sensitivity, and potential autofluorescence. Variants of GFP with altered spectra or brightness expand multiplexing options but require careful channel planning to avoid spectral overlap.

Applications span basic research and biotechnology. GFP-expressing organisms and cells are used for promoter assays, lineage tracing, and real-time monitoring of gene expression. Common examples include GFP-expressing mice, zebrafish, and plants, as well as GFP-tagged proteins to study localization and dynamics. Safety and ethical guidelines govern their development and use, including biosafety containment, genetic stability considerations, and regulatory compliance.
