E2Fproteiineja is a family of transcription factors that play a crucial role in the regulation of the cell cycle, particularly in the transition from the G1 phase to the S phase. These proteins are essential for controlling the expression of genes involved in DNA replication, cell growth, and apoptosis.

The E2F family consists of several members, broadly categorized into activators and repressors. E2F activators, such as E2F1, E2F2, and E2F3, promote the transcription of genes required for DNA synthesis when the cell commits to dividing. They bind to specific DNA sequences known as E2F sites, often located in the promoter regions of target genes. E2F repressors, like DP1, DP2, and DP3, can form heterodimers with E2F activators and, in certain contexts, act to inhibit gene expression.

The activity of E2F proteins is tightly controlled by the retinoblastoma protein (Rb) and its family members (p107, p130). In the G1 phase, Rb binds to E2F, forming a complex that represses the transcription of E2F target genes. As the cell progresses through G1 and prepares for S phase, cyclin-dependent kinases (CDKs) phosphorylate Rb. This phosphorylation causes Rb to release E2F, thereby activating E2F target genes and initiating DNA replication.

Dysregulation of E2F proteins and their regulators is frequently observed in cancer. Aberrant activation of E2F can lead to uncontrolled cell proliferation. Conversely, defects in E2F activity can impair cell cycle progression and contribute to developmental abnormalities. Therefore, the E2F pathway is a significant area of research in molecular biology and cancer therapy.
