ADMEtoxical

ADMEtoxical is an interdisciplinary framework that integrates absorption, distribution, metabolism, and excretion (ADME) with toxicology to evaluate the safety of chemicals across biological systems. The term is used to describe approaches that link pharmacokinetic processes to toxic effects, enabling a mechanistic understanding of how exposure translates into adverse outcomes. While rooted in pharmacology and toxicology, ADMEtoxical is applied in drug development, chemical risk assessment, and environmental health to predict tissue exposure, identify vulnerable compartments, and refine dose selection for preclinical studies.

The framework encompasses toxicokinetics (how organisms process substances) and toxicodynamics (how substances cause effects), within the context of exposure scenarios. Key methods include physiologically based pharmacokinetic (PBPK) modeling, in vitro–in vivo extrapolation (IVIVE), and in silico predictions, combined with experimental data from cell-based assays and animal studies. Data from biomonitoring and human variability are incorporated to improve realism and account for differences in age, sex, genetics, and disease states.

Applications include informing the design of toxicology studies, supporting dose selection, characterizing safety margins, and conducting risk assessments for pharmaceuticals, industrial chemicals, and environmental contaminants. ADMEtoxical analyses support regulatory submissions by providing transparent, mechanistic rationales for exposure limits and by reducing reliance on animal data through alternative methods and efficient screening.

Limitations and challenges include uncertainties in extrapolating across species and populations, variability in ADME processes, and gaps in data. Standardization of models, documentation, and data quality remains a priority, as does ongoing evaluation of model predictivity and acceptance by regulatory authorities.
