Chemical synthesis of oligonucleotides uses solid-phase phosphoramidite chemistry to add nucleotides stepwise to a growing chain. Limitations and considerations include cost, maximum practical length, error rates, and synthesis biases. Safety, biosecurity, and regulatory issues influence the deployment of synthesized sequences, especially for therapeutic or environmental applications. The field continues to advance with improvements in chemistry, assembly methods, and automation that increase throughput and fidelity.
