It is the process of forming a highly ordered, three-dimensional lattice of protein molecules. This ordered structure, known as a protein crystal, is essential for determining the atomic resolution structure of a protein using X-ray crystallography. The ability to obtain a protein crystal is often the bottleneck in the pursuit of a protein's three-dimensional structure.

The principle behind protein crystallization relies on bringing protein molecules together in a controlled manner such that they arrange themselves in a repeating, symmetrical fashion. This is typically achieved by altering the solution conditions of a purified protein sample. Common methods involve carefully adjusting parameters such as protein concentration, pH, temperature, and the presence of precipitating agents like salts (e.g., ammonium sulfate), polymers (e.g., polyethylene glycol), or organic solvents. Additives like ligands or buffers can also play a crucial role in promoting crystal formation.

There are various screening methods used to find suitable crystallization conditions. These include vapor diffusion (hanging drop and sitting drop), microbatch, and dialysis. In vapor diffusion, a small drop containing the protein and precipitant is equilibrated with a larger reservoir solution. Water slowly evaporates from the drop, increasing the protein and precipitant concentration, ideally leading to crystal nucleation and growth.

The success of protein crystallization is highly dependent on the quality and purity of the protein sample. Misfolded or aggregated proteins are generally poor candidates for crystallization. Once suitable crystals are grown, they are harvested and subjected to X-ray diffraction analysis, where the scattering pattern of X-rays passing through the crystal provides the data to computationally reconstruct the protein's atomic structure. This structural information is invaluable for understanding protein function, mechanism of action, and for rational drug design.
