Orkonformational is a term used in structural biology to describe a specific category of protein conformations that are associated with orphan receptors—members of the receptor superfamily that lack a known endogenous ligand. The concept emerged in the early 2010s when crystal structures of several orphan G‑protein‑coupled receptors (GPCRs) revealed distinctive structural motifs that differed from those of ligand‑bound orthologs. Researchers coined the term to emphasize the conformational landscape that these proteins occupy in the absence of a natural agonist.

The hallmark of orkonformational states is a highly flexible extracellular loop network combined with a distinctive packing of the transmembrane helices. Unlike the canonical active or inactive states observed in ligand‑bound receptors, orkonformational structures often display a partially collapsed orthosteric pocket and an extended intracellular cavity that may accommodate allosteric modulators. This arrangement is believed to maintain the receptor in a poised or “preactivated” state, ready for communication with intracellular signaling partners without the need for ligand binding.
