Obenzoylation is the introduction of a benzoyl group onto an oxygen atom of an alcohol or phenol to form a benzoyl ester (R-O-CO-Ph). It is commonly used as a protecting-group strategy in organic synthesis, notable in carbohydrate, nucleoside, and natural-product chemistry.

Typical reagents for O-benzoylation include benzoyl chloride or benzoic anhydride, used with a base such as pyridine, triethylamine, or 4-dimethylaminopyridine (DMAP) to accelerate acyl transfer. Reactions are conducted in aprotic solvents like dichloromethane or pyridine, often at or near room temperature. The choice of reagent, base, and catalyst affects rate and selectivity; phenolic substrates and less hindered alcohols generally benzoylate more readily than hindered ones.

Selectivity depends on steric and electronic factors. Primary and less hindered hydroxyl groups tend to be benzoylated first, and in polyols such as sugars, O-benzoylation is used to protect multiple hydroxyls in defined sequences. Benzoyl groups can also be introduced onto phenols to form aryl benzoates, influencing properties such as solubility and stability.

Deprotection of benzoyl esters is typically achieved by hydrolysis under basic conditions (for example, aqueous NaOH or K2CO3 in methanol or THF) or by transesterification with methoxide. Benzoyl esters are generally more stable than acetates, which can be advantageous during multiple synthetic steps but require harsher conditions for removal.

Advantages of obenzoylation include strong protection and straightforward deprotection, while limitations include slower rates for hindered substrates and the need to remove bulky benzoyl groups at the end of a synthesis.
