Nontoxicity is the absence of harmful effects from a substance under defined conditions. It does not imply universal safety; rather, it describes a lack of detectable toxicity within a specified dose, exposure time, and biological system.

Assessment of non-toxicity typically involves toxicology testing, including in vitro assays that measure cell viability or other health markers, as well as in vivo studies in animals. Regulatory frameworks often require identifying a no observed adverse effect level (NOAEL) or establishing a safe exposure level and margins of safety. Non-toxic conclusions are dose- and context-dependent and may not hold across different routes of exposure or durations.

In pharmacology and drug development, researchers seek candidate substances with non-toxic profiles at therapeutic doses and acceptable safety margins. In biomaterials and consumer products, non-toxicity is a key aspect of biocompatibility. The term is often used in contrast to cytotoxicity, organ-specific toxicity, or systemic toxicity, and is assessed against endpoints such as organ function, blood markers, and reproductive effects. NOAEL and LOAEL (lowest observed adverse effect level) terms are used to describe observed safety thresholds.

Non-toxicity does not guarantee long-term safety; rare or delayed effects may appear with chronic exposure. Species differences and extrapolation uncertainty complicate translating non-toxic findings to humans. Comprehensive risk assessment considers cumulative exposure, vulnerable populations, and real-world use patterns to define appropriate safety margins.
