Matrixtargeted is a term used in biomedical contexts to describe strategies that specifically target components of the extracellular matrix (ECM) or matrix-associated pathways to influence disease progression or improve therapeutic delivery. The term is not universally standardized and is often encountered in combinations like matrix-targeted therapy, matrix-targeted drug delivery, or matrixtargeted imaging. In practice, matrixtargeted approaches aim to exploit ECM features such as abundant matrix proteins (collagen, laminin, fibronectin), glycosaminoglycans (heparan sulfate), or matrix remodeling enzymes (matrix metalloproteinases) to achieve localized effects.

Rationale: In oncology, the tumor microenvironment is rich in ECM and remodeling enzymes, which can impede drug penetration but also offer targets; matrixtargeted strategies seek to accumulate therapeutics at tumor sites or modulate stiffness, porosity, or signaling. In imaging and diagnostics, ECM-binding ligands or probes can enhance contrast in regions of ECM remodeling. In regenerative medicine, matrixtargeted materials aim to direct cell adhesion and tissue regeneration.

Applications: ECM-binding peptides that anchor drugs to collagen-rich tissues; MMP-cleavable linkers that activate prodrugs; nanoparticles decorated with ECM-targeting ligands; imaging agents designed to bind proteoglycans.

Challenges include ECM heterogeneity across tissues, dynamic remodeling, potential off-target effects, and translating matrix-specific effects from model systems to humans.
