Lipofussiinin is a pigment granule that accumulates in the lysosomes of aging cells. The term is sometimes used interchangeably with lipofuscin and can appear in different transliterations or languages as lipofussiinin. Lipofussiinin denotes a heterogeneous mixture of lipid-containing residues of lysosomal digestion, formed from oxidized lipids, cross-linked proteins, and other indigestible cellular debris.

Formation and distribution

The accumulation of lipofussiinin occurs when cellular autophagy and lysosomal degradation become less efficient with age or under oxidative stress. Indigestible residues persist and form granules that accumulate particularly in post-mitotic cells such as neurons, cardiac myocytes, and retinal pigment epithelium cells. Lipofussiinin granules are distributed throughout the cytoplasm and tend to increase in tissues known for long-lived cells.

Characteristics and detection

Under light microscopy, lipofussiinin appears as brownish-yellow granules. The pigment also exhibits autofluorescence, allowing detection by fluorescence microscopy, often without special staining. Electron microscopy reveals dense, membrane-bound granules with complex internal structure. Quantification of lipofussiinin or related autofluorescent signals is used in aging research and studies of cellular lysosomal function.

Biological significance

Lipofussiinin is regarded as a biomarker of cellular aging and lysosomal activity rather than a direct cause of disease. While elevated or focal lipofussiinin accumulation is observed in several aging-related conditions and neurodegenerative contexts, its role is generally considered correlative rather than causative. Ongoing research investigates how lipofussiinin affects cellular physiology and its potential impact on age-associated pathologies.
