Hypervariable is an adjective used in biology and related fields to describe regions or elements that show a high degree of variation across individuals, species, or populations. The term is applied in two broad contexts: genetic sequences that accumulate mutations rapidly, and components of the immune system whose diversity is central to antigen recognition.

In genetics, hypervariable regions (HVRs) include segments of mitochondrial DNA, notably HVR1 and HVR2 in the control region, which mutate relatively quickly and are used in population genetics, forensic analysis, and studies of evolutionary relationships. Hypervariable regions can also refer to variable number tandem repeats (VNTRs) and other repetitive DNA where repeat number varies among individuals. Mechanisms underlying hypervariability include replication slippage, recombination, and selective pressures.

In immunology, hypervariable regions denote portions of antigen receptor genes that determine binding specificity. In antibodies, the hypervariable loops of the variable domains, known as the complementarity-determining regions (CDR1, CDR2, CDR3), are collectively referred to as hypervariable regions. They are generated by somatic recombination and junctional diversity, producing vast repertoires capable of recognizing diverse antigens. T-cell receptors have analogous hypervariable regions as well.

Functional implications include rapid adaptation, immune evasion by pathogens, and extensive diversity in surface proteins. In research and diagnostics, identifying hypervariable regions helps with haplotype inference, forensic typing, and comparative genomics.

Quantifying variability involves metrics such as nucleotide diversity, heterozygosity, and Shannon entropy, and often relies on multiple-sequence alignments to locate highly variable sites.
