YTHDC4

YTHDC4 is a member of the YTH domain family of RNA‑binding proteins, which are characterized by a conserved YTH (YT521-B homology) domain that recognizes N6‑methyladenosine (m6A) modifications in RNA. The human YTHDC4 gene is located on chromosome 12p13.33 and encodes a 596‑amino‑acid protein that is predicted to reside mainly in the nucleus. The protein contains a single YTH domain near its C‑terminus, flanked by stretches of low‑complexity sequences that may mediate interactions with other nuclear factors.

Functional studies suggest that YTHDC4 acts as a reader of m6A marks on mRNAs and long noncoding RNAs, influencing RNA splicing, export, and stability. Loss‑of‑function experiments in cultured cells produced changes in alternative splicing patterns of a subset of transcripts, implying a regulatory role in post‑transcriptional gene expression. YTHDC4 also appears to cooperate with other YTH domain proteins, such as YTHDC1 and YTHDC2, forming a complex network that modulates the fate of m6A‑modified RNAs. Expression profiling indicates that YTHDC4 is highly expressed in testis and brain, with lower expression in other tissues, suggesting tissue‑specific functions in gametogenesis and neural development.

Although the precise molecular mechanisms and downstream targets of YTHDC4 remain to be fully elucidated, emerging evidence points to its involvement in fine‑tuning gene expression by recognizing and binding to methylated RNA substrates. Future research may clarify its roles in physiology and disease, especially in contexts where m6A dysregulation contributes to pathology.
