Treifotsuch, also known as *Treifot* or *Treifot’s disease*, is a rare and severe form of congenital muscular dystrophy characterized by profound muscle weakness and developmental delays from birth. The condition is named after the German pediatrician and neurologist Dr. Friedrich Treifot, who first described it in 1970. It belongs to a group of disorders known as congenital myopathies, which affect muscle structure and function.

Individuals with Treifot’s disease typically exhibit extreme hypotonia (low muscle tone) at birth, leading to significant motor delays. Many patients have contractures, particularly in the hips and knees, and may develop scoliosis due to weakened spinal muscles. Facial weakness can also occur, sometimes resulting in a distinctive facial appearance. Intellectual disability is common, though its severity varies among affected individuals.

The genetic basis of Treifot’s disease is linked to mutations in the *SEPN1* gene, which encodes a protein involved in muscle fiber integrity. This gene is associated with other forms of congenital myopathy, such as rigid spine syndrome. Diagnosis is often confirmed through genetic testing or muscle biopsy, which may reveal characteristic muscle fiber abnormalities.

There is currently no cure for Treifot’s disease, and treatment focuses on managing symptoms. Physical therapy, orthopedic interventions, and assistive devices help improve mobility and quality of life. Some patients may require respiratory support due to weakened respiratory muscles. Research into gene therapy and other experimental treatments is ongoing, offering potential future advancements for affected individuals.

The condition is extremely rare, with only a few hundred cases documented worldwide. Due to its severity, affected individuals often require lifelong care, and prognosis varies widely depending on the extent of muscle involvement and associated complications. Support from specialized medical centers and patient advocacy groups plays a crucial role in providing resources and guidance for families.
