Proline-containing refers to molecules, peptides, or proteins that include at least one proline residue. Proline is a unique amino acid with a cyclic imino group that constrains the backbone, influencing the folding and conformation of the chain in which it resides. This property differentiates proline-containing sequences from those lacking proline and can affect secondary structure and stability.

In proteins, proline is relatively common and is especially abundant in certain structural proteins. Collagen, for example, is highly proline-rich, with repeating Gly-X-Pro or Gly-X-Hyp sequences that contribute to the stability of the collagen triple helix. PRRs often mediate protein–protein interactions by binding domains such as SH3 or WW, typically recognizing proline-rich motifs like PxxP. These interactions influence signaling pathways, cytoskeletal organization, and other cellular processes.
