Matrix-assisted is a descriptive term used across several scientific disciplines to denote methods that rely on a matrix to aid a chemical or physical process. A matrix refers to a substance that interacts with the target analyte to facilitate detection, growth, deposition, or ionization, often by absorbing energy, stabilizing species, or enabling co-crystallization. The concept is general rather than tied to a single technique, and specific implementations vary by field.

In analytical chemistry, the best-known example is matrix-assisted laser desorption/ionization, or MALDI. In MALDI, a small organic molecule (the matrix) is co-crystallized with the sample on a target. When irradiated with a laser, the matrix absorbs energy and transfers it to the analyte, enabling desorption and ionization with relatively gentle fragmentation. This approach is widely used to analyze large biomolecules such as proteins, peptides, and polymers, and it forms the basis of MALDI mass spectrometry.

Other matrix-assisted methods include matrix-assisted techniques in imaging and deposition. Matrix-assisted laser desorption/ionization imaging maps the spatial distribution of molecules in tissue sections or other samples. Matrix-assisted pulsed laser evaporation (MAPLE) uses a matrix to facilitate the transfer of materials to substrates for thin-film fabrication. Matrix-assisted secondary ion mass spectrometry (Matrix-assisted SIMS) enhances ion yields for higher-mass species in surface analysis.

Matrix selection and preparation are critical in matrix-assisted methods, with common matrices chosen to match the analyte’s chemistry and the desired ionization or deposition outcomes. Proper handling minimizes matrix-related background signals and ion suppression, improving reliability and sensitivity.
