Imprintinghäiriöt

Imprintinghäiriöt refers to a group of rare genetic disorders that affect genomic imprinting. Genomic imprinting is an epigenetic phenomenon where certain genes are expressed in a parent-of-origin-specific manner. This means that either the allele inherited from the mother or the allele inherited from the father is silenced, while the other is active. Imprintinghäiriöt occur when this normal silencing mechanism is disrupted, leading to the inappropriate expression or silencing of specific genes.

The most common imprintinghäiriöt are Prader-Willi syndrome (PWS) and Angelman syndrome (AS). Both conditions are caused by abnormalities in the same region of chromosome 15. PWS is typically caused by the deletion of a paternal segment or the inheritance of two maternal copies of chromosome 15. Individuals with PWS often experience intellectual disability, short stature, behavioral problems, and a constant feeling of hunger. AS is usually caused by the deletion of a maternal segment, a mutation in the maternal UBE3A gene, or the inheritance of two paternal copies of chromosome 15. Symptoms of AS include severe intellectual disability, speech impairment, characteristic facial features, and a happy demeanor with frequent laughter.

Other, less common imprintinghäiriöt include Beckwith-Wiedemann syndrome and Silver-Russell syndrome. Beckwith-Wiedemann syndrome is characterized by overgrowth and an increased risk of certain childhood cancers, while Silver-Russell syndrome presents with pre- and postnatal growth restriction. Diagnosis of imprintinghäiriöt is usually made through genetic testing, including methylation analysis and chromosomal microarray. Management involves supportive care and therapies tailored to the specific symptoms and developmental needs of the individual.
