Feeder-free refers to cell culture systems in which cells are maintained and propagated without a feeder layer of supportive cells, such as mouse embryonic fibroblasts or human feeder cells. In feeder-free culture, cells rely on defined extracellular matrix coatings and chemically defined media to provide the signals needed for growth and maintenance.

This approach is widely used for human pluripotent stem cells, including human embryonic stem cells and human induced pluripotent stem cells. Feeder-free methods typically employ defined substrates, such as laminin-521, vitronectin, or other recombinant or synthetic matrices, combined with serum-free, chemically defined media. The goal is to supply essential growth factors and signals without introducing living feeder cells or animal-derived components.

Advantages of feeder-free systems include reduced variability and cross-species contamination, improved reproducibility and scalability, and easier compliance with regulatory and clinical-grade manufacturing requirements. They also facilitate automation and high-throughput workflows by providing more consistent culture conditions.

Challenges and considerations involve the need for careful optimization for each cell line, as some lines may require specific substrates or media formulations for robust growth and pluripotency maintenance. Long-term genetic and epigenetic stability must be monitored, and the cost and quality of defined substrates and media can influence performance. Feeder-free culture has become a standard platform in modern stem cell research and in translational applications seeking defined, xeno-free production of pluripotent cells.
