Desymmetrization

Desymmetrization is a chemical strategy in which a molecule that possesses symmetry is transformed into a less-symmetric, often chiral, product by selectively modifying one of several equivalent sites. By differentiating formerly identical positions, the overall symmetry of the molecule is reduced, frequently introducing a stereocenter.

In practice, desymmetrization targets prochiral or meso substrates and converts them into enantioenriched or diastereomerically enriched products through regioselective and stereoselective transformations. Catalysts—encompassing enzymes, organocatalysts, or transition-metal systems—are used to differentiate between equivalent sites, yielding a single enantiomer or a preferred diastereomer. Desymmetrization can be accomplished under biocatalytic, chemical, or hybrid conditions.

Common approaches include selective functionalization of symmetry-related groups, such as the desymmetrization of meso compounds by regioselective acylation, oxidation, or C–C bond formation. Enzymatic desymmetrization leverages the selectivity of natural catalysts to access chiral products from symmetric precursors, while chemical desymmetrization relies on designed catalysts to achieve high enantioselectivity or diastereoselectivity in a single operation.

Desymmetrization is distinct from symmetry breaking in physics or from kinetic resolution. It directly converts a symmetric substrate into an asymmetrical product in a single step, rather than separating enantiomers of a racemate. It is a valued strategy in the synthesis of enantioenriched pharmaceuticals, natural products, and complex materials, enabling access to chiral architectures from readily available symmetric starting materials.
