Dbf4Drf1
Dbf4Drf1 is a protein encoded by the Dbf4drf1 gene in various eukaryotic organisms. The protein is a regulatory subunit that interfaces with the catalytic Cdc7 protein to form the Dbf4-dependent kinase (DDK) complex. Dbf4Drf1 is characterized by the presence of a N‑terminal regulatory domain rich in acidic residues, a central “D‑box” motif that mediates degradation during the cell cycle, and a C‑terminal domain that binds Cdc7. The DDK complex is conserved across yeast, plants, insects and mammals and plays a critical role in initiating chromosomal DNA replication during the late G1 to S phase transition.
During the S phase, Dbf4Drf1 associates with Cdc7 to phosphorylate components of the pre‑replication complex (pre‑RC).
The Dbf4Drf1 pathway has emerged as a potential target for anti‑cancer therapeutics. Small‑molecule inhibitors that disrupt
