Dbf4Drf1

Dbf4Drf1 is a protein encoded by the Dbf4drf1 gene in various eukaryotic organisms. The protein is a regulatory subunit that interfaces with the catalytic Cdc7 protein to form the Dbf4-dependent kinase (DDK) complex. Dbf4Drf1 is characterized by the presence of a N‑terminal regulatory domain rich in acidic residues, a central “D‑box” motif that mediates degradation during the cell cycle, and a C‑terminal domain that binds Cdc7. The DDK complex is conserved across yeast, plants, insects and mammals and plays a critical role in initiating chromosomal DNA replication during the late G1 to S phase transition.

During the S phase, Dbf4Drf1 associates with Cdc7 to phosphorylate components of the pre‑replication complex (pre‑RC). This phosphorylation activates Mcm2‑7 helicases and other replication initiation factors, allowing the unwinding of DNA and the recruitment of DNA polymerases. In addition to replication, Dbf4Drf1 contributes to the fidelity of chromosome segregation by modulating kinetochore assembly and cohesion maintenance. Loss-of-function mutations in Dbf4drf1 in yeast lead to delayed S phase entry, increased sensitivity to replication stress and chromosome instability. In mammalian cells, Dbf4Drf1 is necessary for maintaining genome integrity during rapid cell division, and its expression is tightly regulated by cyclin‑dependent kinases and the anaphase‑promoting complex.

The Dbf4Drf1 pathway has emerged as a potential target for anti‑cancer therapeutics. Small‑molecule inhibitors that disrupt the Dbf4–Cdc7 interaction are under investigation for their ability to selectively impede DNA replication in tumor cells. Moreover, overexpression of Dbf4Drf1 has been linked to aggressive tumor phenotypes in certain cancers, suggesting its utility as both a biomarker and a therapeutic target. Ongoing research continues to map the regulatory networks that modulate Dbf4Drf1 activity and to explore its roles in development, stress responses and disease.
